DFT studies of the copper active site in AA13 polysaccharide monooxygenase

AA13 polysaccharide monooxygenases (AA13 PMOs) are novel enzymes that break down starch using a copper active site in substrate binding groove on solvent-exposed surface. The structure of the is influenced by residues groove, while crystal Cu(II)-AA13 was damaged photoreduction and lacked two exogenous ligands. We utilized density functional theory (DFT) calculations to obtain insights into presence absence key residue (G89) interferes with distal coordination site. Results show PMOs can exhibit both 6-coordinate 5-coordinate structures depending position G89. features intermediate those AA9 AA10 PMOs, which most abundant well characterized PMO families. In addition, superoxo species has structural parameters halfway between PMOs. relationship intermediates also consistent their evolutionary relationship.
